Efecto del betacaroteno y cobre sobre la producción de embriones in vitro en Jersey

dc.description.abstractLa producción de embriones bovinos in vitro (PIVE) ha permitido explorar un mejor potencial reproductivo de las hembras bovinas, siendo una biotecnología ampliamente usada en la multiplicación de animales con alto potencial productivo. A pesar de todos los avances realizados en los últimos años en PIVE existe una baja eficiencia en los procesos de selección de oocitos ya que tan solo cerca del 40% producen embriones viables luego de ser madurados y fertilizados in vitro (Pfeiffer et al. 2014). Por esto en los últimos años se han venido desarrollando investigaciones enfocadas hacia la aplicación de métodos que favorezcan mejorar la competencia ovocitaria en ganado bovino (Batista et al. 2014; Ribeiro et al. 2014), por esto el objetivo de este estudio fue evaluar el efecto de la aplicación de βcaroteno y cobre en donantes Jersey sobre la producción de embriones in vitro. En este proyecto se seleccionaron hembras bovinas Jersey (n=20), se conformaron dos grupos homogéneos. Al grupo experimental (T) se les aplicó β-caroteno (Betaferol®, Laboratorio Pharvet) a la dosis sugerida por el fabricante (20ml) y Cu (Cobrex®, Laboratorio Pharvet) (10ml) en los días -21 y +35, tomando como día 0 al día del parto. Al grupo control (C) se le aplicó solución fisiológica, a la misma dosis y frecuencia que al grupo T. Previo a la punción folicular (día 45 post parto), se realizó una sincronización de la onda folicular en los animales de ambos grupos. Los oocitos y embriones se cultivaron en todos los procesos de manera individual para cada donante y se llevaron a los procesos de maduración in vitro, fertilización in vitro y cultivo de embriones in vitro de acuerdo con los protocolos del laboratorio comercial. Se registró el semen utilizado ya que este dependió de los requisitos de los clientes de la compañía. En el proceso de búsqueda y selección de los oocitos se evaluó la calidad mediante el uso de la tinción del azul brillante de cresilo para determinar la competencia oocitaria.spa
dc.description.abstractThe production of bovine embryos in vitro (PIVE) has made it possible to explore a better reproductive potential of bovine females, being a biotechnology widely used in the multiplication of animals with high productive potential. Despite all the advances made in recent years in PIVE, there is low efficiency in the oocyte selection processes since only about 40% produce viable embryos after being matured and fertilized in vitro (Pfeiffer et al. 2014). . For this reason, in recent years, research has been carried out focused on the application of methods that favor improving oocyte competence in cattle (Batista et al. 2014; Ribeiro et al. 2014), for this reason the objective of this study was to evaluate the effect of the application of β-carotene and copper in Jersey donors on the production of embryos in vitro. In this project, female Jersey cattle were selected (n=20), two homogeneous groups were formed. To the experimental group (T) β-carotene (Betaferol®, Laboratorio Pharvet) was applied at the dose suggested by the manufacturer (20ml) and Cu (Cobrex®, Laboratorio Pharvet) (10ml) on days -21 and +35, taking the day of delivery as day 0. Physiological solution was applied to the control group (C), at the same dose and frequency as group T. Prior to the follicular puncture (day 45 postpartum), follicular wave synchronization was performed in the animals of both groups. The oocytes and embryos were cultured in all processes individually for each donor and were taken to the in vitro maturation, in vitro fertilization and in vitro embryo culture processes according to the commercial laboratory protocols. The semen used was recorded as it depended on the requirements of the company's clients. In the process of searching and selecting oocytes, quality was evaluated by using cresyl brilliant blue staining to determine oocyte competence.eng
